Hansatech Continuous Excitation Chlorophyll Fluorescence


植物進行光化學的能力是有限的,並且取決於一系列因素,包括環境條件引起的壓力。吸收的光能超過用於光化學的光能時植物必須透過非光化學過程有效地消散能量。這些過程包括熱能的發射和吸收輻射作為較長波長的紅外/遠紅外光能量的重新發射。這種光能的重新發射稱為葉綠素螢光。

儘管整個葉子系統的葉綠素螢光發射太弱而無法用肉眼觀察到,但可以從葉綠素溶液的萃取物中觀察到。葉綠素螢光峰值出現在光譜的紅色區域 (685 nm) 並延伸到紅外線區域約 800 nm。

這些過程中的每一個過程都直接競爭有限的吸收能量,此過程能量利用的任何變化都會導致其他過程的互補變化。使得葉綠素螢光能夠用作快速且可靠的非侵入性光化學探測。

葉綠素螢光儀專門設計用於檢測樣品中的葉綠素螢光發射。有幾種不同類型的葉綠素螢光儀可使用。

◎連續激發葉綠素螢光儀 
葉綠素螢光儀使用紅色 LED 發出的聚焦高強度光來誘導暗適應樣本的快速葉綠素螢光反應。連續系統使用特殊的葉片夾具系統。可為樣品提供暗適應(測量最大光化學效率)、使用高靈敏度光電二極體測量樣品區域並防止環境光洩漏到葉綠素螢光檢測之設計。
